Born in the Mayenne region of France (born in May 2006), Jonathan Do Marcolino made his first steps on a football field with his local youth team at US Laval, where he would play until the age of 11, at which point he joined the Stade Rennais Academy setup. In the 2023/2024 season, he made his debut in the Reserve Team playing in National 3 (French regionalised Fifth Division), and earned the title Best Newcomer at a tournament of the French Academies. In the 2024/2025 season, he made 22 appearances and became a regular in the starting line-up with the Reserves, earning him the privilege of the captain's armband.
His performances didn't go unnoticed, and the club offered him his first professional contract, and the defender signed a three-year-deal on 8th July 2025, before being loaned out to National 1 side, Bourg-en-Bresse soon afterwards.
